In biology, a cell  is defined as the basic membrane-bound unit that contains the fundamental molecules of life and of which all living things are composed. Cells can interact between each other by expressing and secreting different molecules. Under a specific stimuli, cell signaling cascades are activated giving changes to protein activity and expression. Fluorescence immunostaining techniques on cultured cells or individual cells help to follow the protein expression and activity changes by targeting specific antigens with fluorescence-labelled antibodies.

As an individual, cells do not react the same way with a stimulus. Even when using cultured cell lines, some cells may express proteins differently and/or activate them at different levels. The analysis of protein profiles on cultured cells can therefore identify some cell sub-populations. To achieve this, it would be necessary to analyse the whole cell population on a support, under the same conditions. This makes possible the comparison of fluorescence levels among different cells on the sample.
