Extracellular Vesicle Antibody Microarray for Multiplexed Inner and Outer Protein Analysis

A study published in ACS Sensors introduces an innovative method called EVPio (Extracellular Vesicle Inner and Outer Protein Analysis) for multiplexed profiling of proteins in extracellular vesicles (EVs). The research leverages antibody microarrays and the InnoScan 1100 AL confocal microarray scanner to achieve high-throughput, sensitive detection of both surface and internal EV proteins.

The microarray platform enables precise capture of EV subpopulations based on specific surface markers. After fixation, permeabilization and antigen retrieval, oligo-barcoded antibodies and branched DNA amplification enhance detection sensitivity. This approach revealed distinct protein co-expression patterns in EVs from colorectal cancer cell lines, offering new insights for biomarker discovery.
